First, force a check on the disk for corrupt files. NTFS is a pretty good
journalizing filesystem, but it does occasionally get corrupted. Just open
a console window and type "chkntfs /?" for info on how to schedule a check
at next reboot.
Second, might be simple MetaBase corruption. The MetaBase is a special
data file that holds the configuration for IIS. Have you tried to delete
the Website in the IIS Management Console? A simple delete and recreate
might fix it if the MetaBase got whacked.
