I think some people might be a bit confused about whether you mean the
SMTP server on an IIS box, or an actual Exchange box. 

I'm not sure if you're using Exchange 5.5 or 2000 (it's not clear from
the above), but here's the Exchange 5.5 setting that you might well be
after. I'm sure it's changed in Exchange 2000; surely Microsoft at least
TRIED to improve the Exchange Administrator Interface. Lord knows they
couldn't possibly make it WORSE. Anyway, it should be vaguely similar.

Microsoft Exchange Administrator -> <domain> -> Configuration -> 
Connections -> Internet Mail Service -> Routing -> Routing 
Restrictions -> Hosts and Clients with these IP Addresses.

Add the IP addresses or subnets of whichever machines on your network
need to relay mail via that exchange box. If the exchange box is multi-
homed, you can use 'these internal addresses' to open up relaying from
your internal network ONLY.
